I. Background | |
SIRPα(Signal Regulatory Protein α)是一种免疫球蛋白超家族跨膜蛋白,主要表达于 髓系细胞(如巨噬细胞、树突状细胞)表面,其胞外区含三个 Ig 样结构域(D1、D2、 D3),其中 D1 结构域特异性结合 CD47;胞内段含免疫受体酪氨酸抑制基序(ITIM), 可招募 SHP-1/SHP-2 磷酸酶传递抑制信号。SIRPα通过调控吞噬活性维持自身免疫耐受, 防止巨噬细胞误伤正常细胞。
CD47 是一种五次跨膜蛋白,广泛表达于红细胞、血小板及肿瘤细胞表面,其胞外 N 端 IgV 样结构域与 SIRPα结合,被称为“别吃我”(Don’t eat me)信号分子,在红细胞稳 态(防止脾脏巨噬细胞清除年轻红细胞)及肿瘤免疫逃逸中起关键作用。 当 CD47 结合 SIRPα后,SIRPα胞内 ITIM 基序被磷酸化,招募 SHP-1/SHP-2 磷酸酶, 通过去磷酸化作用抑制下游促吞噬信号。 |
