I. Background | |
胰岛素受体(INSR,Insulin Receptor)是调控葡萄糖代谢、细胞生长及能量稳态的核 心跨膜受体。INSR 由两个胞外α亚基(配体结合域)和两个跨膜β亚基(激酶活性域)通过 二硫键连接成四聚体(α2β2)。胰岛素结合α亚基后,触发β亚基自磷酸化并激活其酪氨酸 激酶活性,进而通过 PI3K-AKT 和 MAPK/ERK 信号通路协调代谢与增殖功能。INSR作为代谢与增殖信号的“分子开关”,在糖尿病、癌症及罕见遗传病中具有核心地位。 |
