I. Background | |
整合素αvβ6 是由αv 和β6 亚基组成的异二聚体跨膜受体,属于整合素家族成员,其胞 外域通过识别含 RGD(精氨酸-甘氨酸-天冬氨酸)序列的配体介导细胞与细胞外基质的相 互作用,主要在上皮细胞中低表达,但在组织损伤、纤维化、炎症及恶性肿瘤中显著上调, 成为疾病治疗的潜在靶点。该整合素的主要配体包括细胞外基质蛋白如纤维连接蛋白 (Fibronectin)、层粘连蛋白(Laminin)以及潜活形式的 TGF-β,后者通过与αvβ6 结合被 激活为活性形式,驱动纤维化和免疫抑制微环境形成;此外,αvβ6 还可作为口蹄疫病毒 (FMDV)等病原体的入侵受体。 |
