I. Background | |
CD40 是肿瘤坏死因子受体超家族(TNFRSF)成员,作为免疫调节的关键分子,广泛 参与 B 细胞活化、抗原呈递细胞(APC)成熟及 T 细胞免疫应答。其配体 CD40L(CD154) 主要表达于活化的 T 细胞,通过 CD40-CD40L 互作桥接先天性与适应性免疫,是肿瘤免疫 治疗、自身免疫病及感染性疾病的热门靶点。 CD40 信号转导主要利用 TNF 受体相关因子的衔接蛋白,导致经典和非经典 NFκB 通 路、MAPK、PI3K 和 PLCγ的激活。它们的激活导致特征性下游效应,包括转录激活、细 胞骨架重排和细胞存活。 |
